Training objectives

The student, at the end of the course Laboratory of Oral Microbiology, will know the principles of diagnostic procedures used in microbiology, regarding the isolation and characterization of bacteria and mycetes from the oral cavity. The student will be able to set up a bacterial culture from a specimen obtained from the oral cavity. In addition, the student will be able to measure the microbial load and identify the isolated microorganism, to set up an antibiogram and understand its results, with the aim to identify the adequate drugs for the treatment and eradication of the identified bacteria.

Prerequisites

To attend profitably the course Laboratory of Oral Microbiology, the student must have knowledge of Microbiology, regarding the structure, classification, metabolism and replicative cycle of the human pathogens. In addition, the student must also have knowledge of the bacterial media and culture conditions.

Course programme

Basic aspects of infectious diseases diagnosis.
Diagnostic procedures used in microbiology.
Sampling methodologies for oral microorganisms and their culture isolation.
Plating techniques for isolation of bacteria and mycetes in different basic, enriched and selective culture media.
Bacteriological differential staining (Gram, Ziehl Nielsen).
Single colony isolation and typization of isolated microorganism (biochemical typization by API system).
Drug resistances and antibiogram: antibiotic susceptibility test; Kirby-Bauer assay.
Immunofluorescence reactions applied to oral microbiota analysis.
Analytical reading of the microbiological tests.

Didactic methods

The course develops in one intensive week, during which the daily frequency of the laboratory is mandatory (about 4 hours/Day). The course is both theoretical and practical: it starts with lectures followed by practice exercises, which cover most of the activities. Each student will have his/her own material, to put into practice what treated in the theoretical lecture. The student will perform the sampling from his/her oral cavity, the seeding of microorganisms by plating on selective media, the microscopic observation after Gram staining, the biochemical typization by API systems, and the antibiotic susceptibility tests.

Learning assessment procedures

Oral test, including about 3 questions on different topics, in a total time of about 20-30 minutes, in which the exposition precision and clarity, the connection ability and the communication skills will be evaluated.
